FUTURE STANDARD OF CARE FOR THE TREATMENT OF X-LINKED RETINOSCHISIS
What Are IRDs?
Inherited Retinal Diseases (IRDs) are a diverse group of rare genetic disorders that lead to progressive degeneration of the retina—the light-sensitive tissue at the back of the eye. These conditions can result in significant vision impairment and, in many cases, blindness.
Prevalence and Global Impact
IRDs collectively affect approximately 1 in 2,000 to 1 in 3,500 individuals worldwide, translating to an estimated 5.5 million people globally living with these conditions PNAS.
Genetic Diversity
- Over 250 genes have been identified as causative for IRDs, with many more likely undiscovered Lions Eye Institute.
- These conditions exhibit genetic heterogeneity, meaning that a single disease phenotype can result from mutations in different genes, complicating diagnosis and treatment development J&J Medical Cloud.
Diagnosis Challenges
- Diagnosing IRDs can be complex due to their genetic diversity and phenotypic overlap with other ocular conditions.
- Genetic testing is often required to confirm a diagnosis, but access to comprehensive genetic screening may be limited, especially in underserved regions PMC.
Impact on Quality of Life
- Studies have shown that patients with IRDs report lower quality of life scores, particularly in areas related to independent living, sensory function, and social relationships PMC.
- According to the Centers for Disease Control and Prevention (CDC), the estimated economic burden of vision loss and blindness in the United States is $134.2 billion annually.
Ophthalmology
Unmet Medical Need
- Currently, most IRDs have no approved drug treatments, leaving patients with limited options.
- The lack of effective therapies underscores a substantial unmet medical need, highlighting the importance of ongoing research and development in this field PMC.

Approach
RareSight has deep expertise in pharmacology, drug discovery, clinical strategy and commercialization within ophthalmology and rare disease. We are exploring new treatment approaches for rare inherited retinal diseases, focusing on areas where no approved therapies exist. Partnering with physicians and drug delivery innovators, we are working to translate scientific insight, real-world evidence and clinical expertise into future treatment solutions.
Our goal is to design therapies that are not only effective but also practical in a pediatric setting with the mission of preventing children from losing vision early in life and helping them maintain sight throughout their lives.
Team
Carmen Caricchio
Founder & CEO
Carmen Caricchio is the CEO and Founder of RareSight, bringing more than 20 years of experience in the ophthalmic industry spanning product development, commercialization and strategic communications. Throughout the course of her career, she has brought more than 30 market-leading technologies in cornea, glaucoma, and retina into clinical practice.
Since 2018, Carmen has advised ophthalmic companies—from early-stage start-ups to public entities—on commercialization strategy, market evaluation, product development, clinical adoption, investor engagement and go-to-market planning. She also serves as an Expert-in-Residence at UCSF Innovation Ventures for the Department of Ophthalmology, where she evaluates new inventions and accelerates spin-out opportunities.
Prior to this, Carmen specialized in public relations, strategic communications and public affairs for the MedTech industry. She was the Founder/Principal of the award-winning CoActive Communications with offices in San Francisco, New York City and Sydney, Australia. During its 15-year span, her agency maintained a large portfolio of ophthalmic start-up clients, most of which were acquired by major industry strategics, due in part, to her agency’s steadfast media engagement, publication and podium strategies. Carmen was introduced to ophthalmology in 2002 as head of the West Coast office of Dorland Global, then the largest independently owned healthcare advertising agency in the U.S. Earlier in her career, Carmen headed up MCS WEST, the MedTech practice for New Jersey-based, MCS Health.

PolyActiva and RareSight Form Strategic Collaboration to Develop Breakthrough Treatments for Rare Pediatric Retinal Diseases
FORT WORTH, Texas, MELBOURNE, Australia, and MENLO PARK, California — November 5, 2025 — PolyActiva, a clinical-stage biopharmaceutical company pioneering a novel drug delivery technology to improve outcomes for patients with ocular conditions, and RareSight, Inc., an ophthalmology-focused life sciences company committed to advancing therapies for inherited retinal diseases (IRDs), today announced a strategic collaboration to develop first-in-class therapies for rare pediatric retinal diseases that currently have no approved drug treatments.
The research collaboration agreement, leveraging PolyActiva’s proprietary PREZIA™ platform, will offer first-of-its-kind, new chemical entity (NCE)-eligible pro-drug candidates to people affected by blinding eye conditions. Administered intravitreally, the approach will deliver sustained-release, zero-order treatment directly to the retina, requiring no patient administration—a key benefit for children and their caregivers.
